KIST Finds Low Doses of Toxic Quinolinic Acid Wake the Brain's 'Cleaning Cells' in Mice

한국과학기술연구원(KIST)의 류훈·이현범 연구팀은 미국 보스턴대 의과대학 이정희 교수팀과의 공동 연구에서, 독성 물질로 알려진 퀴놀린산이 낮은 농도에서는 뇌의 청소 세포를 깨운다는 사실을 확인했습니다. 퀴놀린산은 농도가 높아지면 신경세포를 손상시키는 물질로 알려져 있습니다. 그런데 낮은 농도에서는 정반대로, 뇌의 청소 세포인 미세아교세포의 노폐물 제거 능력을 높이는 것으로 나타났습니다. 세포실험에서 저농도 퀴놀린산이 미세아교세포를 자극하자 세포막의 지방 성분인 인지질을 만드는 경로의 핵심 효소가 활성화됐고, 이때 만들어진 ’PE 인지질’이 자가포식에 관여하는 단백질 GABARAP과 결합했습니다. 알츠하이머병 생쥐 모델의 뇌에 저농도 퀴놀린산을 투여하자 며칠 만에 해마의 아밀로이드 베타 플라크가 유의하게 줄었고, 단기·장기 기억 능력은 정상 생쥐 수준으로 좋아졌습니다. 연구 결과는 국제학술지 Signal Transduction and Targeted Therapy에 실렸으며, 연구팀은 제약·바이오 기업과 함께 전임상 검증을 추진할 계획입니다.
Translation
In joint research with the team of Professor 이정희 at Boston University's medical school, a team at the Korea Institute of Science and Technology (KIST) led by 류훈 and 이현범 confirmed that quinolinic acid, known as a toxic substance, wakes up the brain's cleaning cells at low concentrations. Quinolinic acid is known to damage nerve cells when its concentration rises. At low concentrations, however, it was found to do the opposite and raise the waste-clearing ability of microglia, the brain's cleaning cells. In cell experiments, when low-dose quinolinic acid stimulated microglia, a key enzyme in the pathway that makes phospholipids, the fat component of cell membranes, was activated, and the 'PE phospholipid' produced then bound to GABARAP, a protein involved in autophagy. When low-dose quinolinic acid was administered into the brains of Alzheimer's model mice, amyloid beta plaques in the hippocampus fell significantly within days and short- and long-term memory improved to the level of normal mice. The results were published in the international journal Signal Transduction and Targeted Therapy, and the team plans to pursue preclinical validation with pharmaceutical and biotech companies.
